The Octet system is fully controlled by software that simplifies assay setup, data acquisition, and analysis. An Experiment Wizard assists the operator through each step via simple prompts, facilitating experimental setup, and results are displayed in real time during data collection.

Data analysis can be performed either at the instrument or on a remote computer, streamlining the workflow to increase efficiency and minimize operator time. For each experiment, a directory folder is created, in which raw data files for each sensor will be saved. Raw data is always accessible, if archiving is desired.
Users can save operator time and reduce the potential for error by saving experimental method files in advance and recalling at runtime. The software also supports importing tab-delimited text or Notepad files for sensor and sample ID, and concentration/sample/buffer information.

The software’s programmable dilution factors for unknown samples simplify quantitation during data analysis, while click-and-drag functions simplify assigning well types to multiple wells (e.g., standards, unknowns or controls).
Real-Time Data Acquisition
The Runtime binding view window shows plates' experimental status, sample progress and elapsed time, enabling rapid evaluation of raw data for expected trends and monitoring binding progress. A highly interactive interface makes customizing the assay window easy and intuitive. The user can automate and speed up assay development by setting a threshold value for the binding signal in any step in the experiment. Reference-subtracted data can be viewed in real time — even while data is being acquired. For a summary of the major features in data acquisition features in version 6.3, see the table below.
Data Analysis
The data analysis options are extremely powerful and flexible. An entire experiment, or individual files (sensor raw data) from single or multiple experiments can be selected, to optimize the data analysis and workflow. Users can apply a single set of standards to any number of sample microplates, or, apply calibration for unknowns within each microplate. For kinetic screening and characterization, the software includes 2:1 Heterogeneous ligand interaction analysis. Due to unique circumstances in some binding systems, data may be influenced by mass transport limitation. The Mass Transport fit in the Octet data analysis software enables you to analyze such data to extract true binding constants. Flexible Data Management Options
After analysis, a complete data report can be assembled from the various charts and tables according to your requirements, then exported as a Microsoft Excel document for easy lab notebook layout and subsequent analysis. Graphs can be exported as jpeg files at any time. Additionally, ForteBio recognizes scientists in life science organizations need to handle low to high volumes of data generated from routine screening on the Octet systems. We are actively working in collaboration with data management software providers such as IDBS to enable users to easily transfer data from the Octet software to databases such as the ActivityBase suite.
