Octet System
The Octet System performs label-free quantitation and kinetics analysis of antibodies and other proteins in real time. In comparison to other label-free technologies, the Octet System brings the detection surface directly to a 96-well sample plate, eliminating the need for microfluidics. This “dip and read” format is configured for eight-channel parallel analysis, enabling operators to increase throughput without increasing budget or headcount. Workflow delays are eliminated by the system’s crude sample compatibility. Samples can be taken directly from bioreactors or periplasmic preps without interference.
To meet the requirements of research, development and bioprocessing workflows, the Octet System is available in two configurations:
Octet Q System
The Octet Q instrument rapidly measures the concentrations of antibodies and other proteins in solution. The system offers the speed of ELISA assays and the accuracy of HPLC. Ninety-six samples can be processed in as little as 32 minutes. Analytes in cell culture media and crude lysates solutions can be measured accurately without serially diluting samples or centrifugation. Raw data is displayed in real time during sample analysis. If your workflow requires timely protein concentration data, choose the Octet Q System.
Octet QK System
To obtain the maximum benefit of label-free, real-time detection, the Octet QK expands your capabilities to perform full kinetic characterization and screening of protein-protein interactions, including kobs, ka, kd, and KD. Choose the Octet QK System for rapid, benchtop protein quantitation and kinetics.
Key Benefits
- Ease of Use
No microfluicdics translates into virtually no maintenance and a more rapid learning curve.
- Minimal Interference
Because the Octet System only detects binding at the sensor surface, there is minimal interference from biological sample media.
- Automated
The Octet System is automated to perform multi-step experimental protocols and complete data analysis. The system runs up to eight samples in parallel, and up to 96 samples in unattended operation.
Octet Software, For Simple, Real-time Detection of Molecular Interactions
The Octet System is fully controlled by software that simplifies assay set-up, data acquisition, and analysis. An Experiment Wizard, which assists the operator through each step with simple prompts, facilitates experimental set-up and results are displayed in real time during data collection.
Data analysis can be performed either at the instrument or a remote computer, streamlining the workflow to increase efficiency and minimize operator time. For each experiment, a directory folder is defined where raw data files for each sensor will be located. You always have access to the raw data for archiving if required.
Pre-analysis Set-up
Save operator time and reduce the potential for error, by saving experimental method files in advance and recalling at runtime or for future experiments. Users also have the option of importing tab-delimited text or Notepad files for sensor and sample ID, concentration, and sample/buffer information.

Octet Software setup is as simple as 1-2-3.
The software’s programmable dilution factors for unknown samples simplify quantitation during data analysis, while click-and-drag functions simplify assignment of well types to multiple wells (e.g., standards, unknowns or controls).
Real-Time Data Acquisition
The full screen display shows plate experimental status, sample progress and elapsed time enabling quick evaluation of raw data for expected trends and to monitor the progress of antibody production. For kinetic experiments, if binding rates are other than expected, the operator has the option of accelerating to the next step, or delaying the transition.

Data Analysis
The data analysis options are extremely powerful and flexible. Select an entire experiment, or individual files (sensor raw data) from single or multiple experiments, to optimize the analysis and workflow. Each Octet System comes complete with Origin Software and a single-user license.
Flexible Data Management Options
After analysis, a complete data report can be assembled from the various charts and tables according to your requirements, then exported as a Microsoft® Word or Microsoft Excel document for easy lab notebook layout and subsequent analysis. Graphs can be exported as a jpeg file at any time.
