Octet QKe and QK System

The Octet QKe SystemThe Octet QKe and QK systems perform label-free quantitation of antibodies, proteins, peptides, DNA and other biomolecules and provide kinetic characterization of biomolecular binding interactions. Octet systems bring the detection surface directly into a 96-well sample plate in a simple Dip and Read format, eliminating the need for microfluidics and the instruments are configured for eight-channel parallel analysis, enabling operators to increase throughput without increasing budget or headcount. Workflow delays are eliminated by the system’s compatibility with crude samples can be taken directly from bioreactors or periplasmic preparations without interference.

Octet Qke System

The Octet QKe instrument is the newest addition to the Octet family of real-time, label-free biomolecular interaction analysis instruments. It is an enhanced performance version of the first-generation Octet QK instrument. Two acquisition rate settings and a higher-performance spectrometer combine to provide greater sensitivity and wider dynamic range for quantitation and kinetic analysis. Additionally biosensor re-racking provides enhanced flexibility and operational cost savings.

The Octet QKe has the same footprint as the Octet QK and Octet RED systems and has one sample plate position.  The Octet QKe instrument contains the same high performance spectrometer used in the Octet RED system. In addition, new Octet software version 6.3 installed on the Octet QKe provides a High Sensitivity acquisition rate setting for both quantitation and kinetics assays, allowing measurement of lower ranges of concentrations and smaller molecular weight analytes than possible using the Octet QK.

The Standard acquisition rate of 0.6 Hz with 5 averages per data point (1 data point per 1.6 seconds) is the default setting on the Octet QKe and equivalent to that on the Octet QK. The High Sensitivity acquisition rate of 0.3 Hz (1 data point per 3.3 seconds) allows the software to perform 40 averages per data point to reduce noise and enhance signal to noise ratios, and gives the Octet QKe enhanced sensitivity for quantitation of proteins.

Kinetic analysis of proteins and peptides benefits from the Octet QKe enhancements over the Octet QK in two ways:

  • Lower molecular weight proteins and peptides can be detected and analyzed on the Octet QKe.
  • The binding data for proteins or peptides at different concentrations can be better resolved on the Octet QKe, providing reliable measurement of kinetic constants.

In comparison, the Octet QK384 provides equivalent performance to the Octet QKe at greater throughput. The Octet RED and Octet RED384 provide the best performance among the Octet family of instruments for quantitation and kinetic analysis, with the broadest dynamic range, greatest sensitivity, ability to measure small molecule:protein binding and ability to measure fast sub-nM binding interactions.

Biosensor Re-racking

The Octet QKe system provides the ability to re-rack biosensors at the end of an experiment. Biosensor re-racking provides enhanced flexibility in loading ligands on biosensors and reduces operational costs when biosensors are loaded with precious target reagents. Combining lower costs with increased flexibility and superior data quality, the Octet QKe provides a value-added alternative to other label-free instruments for your assays.

The Octet QK SystemThe Octet QK is a first-generation Octet system that performs label-free quantitation of proteins and peptides and kinetic characterization of various biomolecular binding interactions.

The Octet QK is automated to perform multi-step experimental protocols and complete data analysis. The system runs up to eight samples in parallel, and up to 96 samples in unattended operation. The Octet QK is a versatile instrument that provides concentration data for antibodies and other proteins in crude lysates, untreated supernates, periplasmic fractions and other complex samples and analyzes protein-protein interactions to provide kobs, ka, kd and KD information. The Octet QK has one acquisition speed setting of 0.6 Hz (1 data point per 1.6 seconds) with 5 averages per data point.
