Octet Instrument Family

ForteBio’s Octet family of instruments and related consumables provides scientists with label-free, real-time information about biomolecular interactions — particularly for proteins, peptides, DNA, RNA, and small molecules. Any user, including non-experts, can perform detection and analysis quickly, easily, and affordably.

Octet RED and Octet

Key Benefits

  • Label-Free Detection
Binding interaction at the biosensor surface does not require the use of detection labels. There are no labeling steps before the analysis, and there is no interference from fluorescent or chromogenic tags.
  • Real-Time Results
Protein binding is continuously measured at the biosensor surface throughout the interaction. Raw data are displayed in real time and the rapid analysis fits with process workflow.
  • Minimal Interference
Binding is only detected at the sensor surface, so there is minimal interference from biological sample media. Proteins can be assayed in crude mixtures (cell lysates or hybridoma sups) or in DMSO (< x%) without interference.
  • Higher Throughput
Assay up to eight samples in parallel, and up to 96 samples in unattended operation. The systems are automated, simplifying the performance of multi-step experimental protocols and complete data analysis.

Life science research, drug discovery, and process development applications include:

KINETIC MODE MEASUREMENTS

QUANTITATION MODE MEASUREMENTS

Kinetics—how specific, fast and tight
are the interactions?

  • Measure association rates — ka
  • Measure dissociation rates — kd
  • Determine affinity constants — Kd
  • Simple yes/no binding

Quantitation—what is the sample concentration?

  • Human IgG quantitation for antibody titer determination or cell line optimization
  • Mouse IgG quantitation for research
  • Custom quantitation assays based on a protein of interest

Options: Octet System or Octet RED System

The original Octet System provides detection and analysis for a broad range of interactions ideal for antibody therapeutic development and production. With higher sensitivity and more rapid data acquisition, the newly launched Octet RED System has extended detection capabilities. It can detect molecules of lower molecular weight, for comprehensive kinetic characterization of small molecules and peptides. Both systems use BioLayer Interferometry (BLI) technology to enable real-time analysis of biomolecular interactions in a flexible, easy-to-use format. BLI-based systems have no microfluidics, so there is virtually no maintenance, and they are ideal for on-demand analysis by multiple users.
