  • Kinetic analysis

    The Octet K2 System monitors binding events in real time to calculate on rates (ka), off rates (kd), and dissociation constants (KD). The system's two channels can be used to measure samples independently or in tandem, pairing the sample read with a dedicated reference. Samples are loaded in standard microplates, eliminating fluidic line maintenance and expanding the number of samples that can be run in an assay. With predefined templates to follow in Octet data acquisition software, the system streamlines setup prior to running an assay and minimizes training needs. The software then gives users a range of parameters and metrics for analyzing acquired data in a powerful yet intuitive software interface.

In addition, ELISA may not provide the information required for the confident understanding of a drug molecule’s MOA. Scientific Poster Quantitation of Residual Protein A and MabSelect SuRe using a “Dip and Read” Assay

Detection of residual Protein A in product purified using a Protein A, or similar column matrix such as MabSelect SuRe™, is a critical quality control step in antibody therapeutic development and production. During the IgG purification process, Protein A can leach from the purification column and co‐elute with the IgG. This contaminant in the therapeutic mixture can cause adverse reactions in patients and thus must be detected and minimized. The Octet family of instruments utilizes a unique label‐free “dip and read” detection technology to quantify biomolecules in solution. ForteBio has developed a new kit for use on this system to detect and quantify residual Protein A. The new Dip and Read™ Residual Protein A Detection Kit contains biosensors and all the reagents required to quantitate Protein A and MabSelect SuRe. The data presented here demonstrates that the Octet system can accurately and precisely detect 100 pg/mL Protein A or MabSelect SuRe in bioprocess samples.

The flexibility and throughput of ForteBio’s Octet system lends itself readily to a variety of screening applications which will be discussed in detail. Clonal screening and selection based on protein expression levels can be an important step in antibody development. Using Protein-A Biosensors, the Octet System can screen 96 samples in 30 minutes to provide accurate quantitation from crude lysates or media samples. Dissociation-rate screening to rank protein affi nities is critical to the development of therapeutic antibodies. Using fi ve-minute off -rates as a selection criterion, 96 samples can be screened against a biotinylated target in little over an hour. Affinity screening on Octet is convenient and rapid and can provide accurate determinations of protein kinetics with a minimum of development time. Using Octet to screen lysates against a series of amine-coupled targets in a multiplexing strategy further accelerates the selection of potential drug candidates.
